Contents
Per 5 mg/50 mg tab Amlodipine besilate 5 mg, losartan K 50 mg. Per 5 mg/100 mg tab Amlodipine besilate 5 mg, losartan K 100 mg
Indications/Uses
Mild to moderate HTN in case of inadequate control w/ monotherapy.
Dosage/Direction for Use
Initially 1 tab daily, increased to 2 tab daily if necessary.
Administration
May be taken with or without food.
Contraindications
Special Precautions
Amlodipine & losartan: Hepatic impairment, hypotension. Amlodipine: Patients w/ poor cardiac reserve, heart failure. Do not use in cardiogenic shock, patients who have recently suffered MI, or in acute unstable angina. Not for the treatment of chronic stable angina. Increased risk of developing heart failure in patients w/ severe aortic stenosis. Exacerbation of angina in sudden w/drawal. Discontinue use in ischaemic pain following administration. Losartan: Renal artery stenosis or renal impairment. Monitor serum K conc in elderly. Avoid concomitant use w/ K-sparing diuretics. Losartan: Pregnancy & lactation.
Adverse Reactions
Dizziness, hypotension. Amlodipine: Flushing, headache, peripheral edema, tachycardia, palpitations; nausea & other GI disturbances, increased micturition frequency, lethargy, eye pain & mental depression; rashes (including erythema multiforme), fever, abnormalities in liver function, including cholestasis due to hyperplasia, myalgia, tremor, impotence. Losartan: Renal function impairment, hyperkalemia, myalgia; resp tract disorders, back pain, GI disturbances, fatigue & neutropenia.
Drug Interactions
Amlodipine: Enhanced antihypertensive effects w/ β-blocker; aldesleukin & antipsychotics that can cause hypotension. May modify insulin & glucose responses when receiving antidiabetic treatment. Other CYP3A4 metabolized drugs eg, quinidine. Enzymes inducers (eg, carbamazepine, phenytoin, & rifampicin) & inhibitors (eg, cimetidine, erythromycin & HIV-PIs). Losartan: Potentiated antihypertensive effects w/ drugs or other agents that lower BP. Additive hyperkalemic effect w/ K supplements, K-sparing diuretics & other drugs causing hyperkalemia. Increased risk of renal impairment w/ NSAIDs. Drugs affecting CYP450 isoenzymes.
